Serum Bilirubin Test
Method: Waller
method
Principle: Serum + Sulfanilic acid + sodium
nitrite Reaction Color complex. Color is measured by a
colorimeter/spectrophotometer.
Articles:
1. Colorimeter/spectrophotometer
2. Test tubes with rack
3. Micropipette
4. Reagents:
a. Sulfanilic acid
b. Sodium nitrate acid
c. Sample: Serum/Plasma
5. Marker, Tissue paper, Cotton etc.
Procedure:
1. Working reagent = 0 ml - 1ml
2. Bland reagent = 1 ml - 0ml
3. Serum = 100 µl - 100µl
Mix and allow to stand for 10 minutes protected frpm light.
Measure absorbance of sample against the sample blank by colorimeter /spectrphotometer.

Test of OD * 12.5
= mg/dl
Normal
range: 0.3 to 1.2 mg/dl
